Not having played Mannpower in a while, just looking at the powerups draws me to a King/Reflect combo for Medic. Obnoxious support from King + Medic's natural healing abilities, coupled with backlash to adversaries from Reflect to give Medic some protection from the occasional flanker. Not an immediate puncher, but more of a long term survival combo.

 

Alternatively, swap out either powerup for Supernova and save the blast for right before an uber.

 

Edit: Not sure how powerups work with ubercharges, but it occurs to me that if you keep your powerup effects while ubercharged, double Medic uber chains could be absolute hell. Stack Plague (and Reflect or Resistance if we're talking combos) onto both of them, then go ubersawing to your heart's content while chaining ubers back and forth.
